Deficienza dell’isoforma 1 del carrier mitocondriale di aspartato/glutammato: meccanismi patogenetici e analisi mutazionale

Il deficit da AGC1 è una malattia genetica che provoca gravissimi difetti neurologici e limita notevolmente lo sviluppo muscolare e le funzioni cerebrali. Uno dei dati clinici più evidenti della malattia è la diminuzione del contenuto di mielina (la guaina che riveste le cellule nervose) nel cervello. Abbiamo identificato come causa della malattia un difetto nel gene della proteina AGC, essenziale per trasportare due tipi di aminoacidi (i mattoni che formano le proteine) nelle cellule nervose e, più precisamente, all’interno dei mitocondri (le centrali energetiche delle cellule). Poiché attualmente non ci sono cure per il deficit da AGC1, il nostro laboratorio sta studiando nel dettaglio in modelli cellulari l’effetto del difetto della proteina e il suo ruolo nell’insorgenza della malattia. Questo permetterà di approfondire i meccanismi che impediscono la corretta sintesi della mielina nel cervello quando AGC1 è inattiva e aiuterà, inoltre, a studiare nuovi approcci terapeutici per alleviare o curare i sintomi della malattia.
