Ruolo del ferro e dei mitocondri nella patogenesi della Neurodegenerazione associata a pantotenato chinasi (PKAN): sviluppo di nuovi modelli cellulari neuronali e analisi di un modello di malattia murino

La neurodegenerazione associata a difetti della pantotenatokinasi (Pkan) è la più frequente tra le rare malattie neurodegenerative caratterizzate da accumulo di ferro nel cervello. Al momento, il ruolo del ferro nello sviluppo della malattia e i meccanismi che ne provocano il sovraccarico nel cervello sono oscuri. Quello che si sa è che la malattia è causata dall’alterazione di una proteina - Pank2 - presente nei mitocondri, le centrali energetiche della cellula. Il nostro gruppo di ricerca è al lavoro per chiarire il ruolo del ferro, di Pank2 e dei mitocondri nella manifestazione della patologia. Il nostro progetto di ricerca mira a sviluppare nuovi modelli cellulari il cui comportamento sia il più possibile simile a quello delle cellule nervose colpite dalla malattia, tappa essenziale per sviluppare approcci terapeutici adeguati. Per farlo utilizzeremo cellule staminali derivate dal follicolo pilifero di pazienti e riprogrammate per divenire cellule nervose e cellule umane in cui il gene Pank2 è reso inattivo. Inoltre, analizzeremo l'attività dei mitocondri nel modello animale della malattia.
